چکیده

The early, de-embryonated chick blastoderm, cultured in vitro, closely simulates a phased culture of erythroid cells. This fact permits the study of hemoglobin synthesis from colorless erythroblast precursor cells to fully hemoglobinated erythrocytes. When b-aminolevulinic acid is added to colorless blastoderms, thereby by-passing &aminolevulinic acid synthetase, copious amounts of porphyrins, and presumably of heme, are formed. This result indicates that in the erythroid precursor cells of the early blastoderms heme synthesis is limited by the activity of &aminolevulinic acid synthetase. The addition of Caminolevulinate also results in an increase in globin synthesis and hemoglobin formation. The enhancing effect of b-aminolevulinic acid on hemoglobin formation is not abolished by actinomycin D but is prevented by puromycin. This result suggests that &aminolevulinic acid forms heme which stimulates globin synthesis at the ribosome level. It is conjectured that heme may be necessary for the appropriate folding of the globin polypeptide in the completion of its synthesis. The hypothesis is proposed that the formation of d-aminovulinic acid synthetase, which is under repressor control, is the limiting and controlling reaction in the formation of hemoglobin. Such a hypothesis would explain the fact that no free globin is formed and that monomeric globin and heme are formed in a 1:l ratio.

Studies of murine erythroid cell development. Synthesis of heme and hemoglobin

Techniques of cell separation were used to isolate murine erythroid precursors at different states of maturation. Cells were studied before and after short-term incubation in the presence or absence of erythropoietin. Complementary results were obtained by direct examination of the cell fractions and by the short-term culture experiments. Indices of heme synthesis, including incorporation of 59...

delta-Aminolevulinic acid synthetase in erythroblasts of patients with pyridoxine-responsive anemia. Hypercatabolism caused by the increased susceptibility to the controlling protease.

Properties of delta-aminolevulinic acid synthetase in erythroblasts of patients with pyridoxine-responsive anemia were investigated with special reference to the protease in mitochondria of erythroblasts. delta-Aminolevulinic acid synthetase activity in erythroblasts of patients with this disease before treatment was extremely decreased, whereas it gradually increased in parallel with the impro...

Erythropoietic activity of steroid metabolites in mice.

The per cent Fe(59) incorporation into circulating erythrocytes as an indicator for new hemoglobin production was used in the polycythemic exhypoxic mouse system to study the effects of certain steroid hormone metabolites on erythropiesis. Enhanced Fe(59) incorporation was observed after the administration of several metabolites with a 5beta-H configuration, while those with a 5alpha-H configur...

Enzyme and nucleic acid formation during synchronous growth of Rhodopseudomonas spheroides.

The synthesis of various cell components was examined during the anaerobic photosynthetic growth of synchronous populations of Rhodopseudomonas spheroides. Net deoxyribonucleic acid (DNA), ribonucleic acid (RNA), and protein increased continuously as did the rate of incorporation of radioactive precursors into protein. The rates of incorporation of radioactive precursors into RNA and DNA were m...

The present study was conducted to evaluate the ameliorating effect of 5-aminolevulinic acid (ALA) on physiological and biochemical changes of induced NaCl-salinity stress on in vitro shoot and callus cultured of red-fleshed apple. Shoot and callus segments of the red-flesh apple of Local Iranian genotype were cultured on MS medium containing different levels of NaCl (0, 30, 60 and 90 mM) and A...
